Combinatrial treatment of anti-High Mobility Group Box-1 monoclonal antibody and epothilone B improves functional recovery after spinal cord contusion injury

چکیده

Spinal cord injury (SCI) causes motor and sensory deficits is currently considered an incurable disease. We have previously reported that administration of anti-High Mobility Group Box-1 monoclonal antibody (anti-HMGB1 mAb) preserved lesion area improved locomotion recovery in mouse model SCI. In order to further enhance the recovery, we here examined combinatorial treatment anti-HMGB1 mAb epothilone B (Epo B), which has been promote axon regeneration. This significantly increased hindlimb movement compared with alone, although Epo alone failed increase functional recovery. These results are agreement was able decrease spreading surviving neuron numbers around lesion, whereas facilitated outgrowth only combination mAb, suggesting mAb-dependent tissue preservation necessary for exhibit its therapeutic effect. Taken together, can be as a novel clinically applicable strategy

Exogenous high-mobility group box 1 improves myocardial recovery after acute global ischemia/reperfusion injury.

BACKGROUND High-mobility group box 1 (HMGB1) is a mediator of inflammation with dose-dependent effects. In the setting of regional myocardial infarction, a high-dose HMGB1 treatment decreases myocardial function, whereas low-dose HMGB1 improves function; however, it is unknown what role HMGB1 has in the setting of global ischemia/reperfusion (I/R) injury. We hypothesized that a low-dose HMGB1 t...

Thermostabilized chondroitinase ABC Promotes Neuroprotection after Contusion Spinal Cord Injury

Background: Chondroitinase ABC (cABC), due to its loosening impact on the extracellular matrix scaffold, has been used to enhance regeneration of injured axonal tracts after spinal cord injury (SCI). However, cABC thermal instability at physiological temperature has limited its clinical application. The disaccharide trehalose has been shown to increase the stability of cABC in body temperature....

Therapeutic potential of an anti-high mobility group box-1 monoclonal antibody in epilepsy.

Brain inflammation is a major factor in epilepsy, and the high mobility group box-1 (HMGB1) protein is known to contribute significantly to the generation of seizures. Here, we investigated the therapeutic potential of an anti-HMGB1 monoclonal antibody (mAb) in epilepsy. anti-HMGB1 mAb attenuated both acute seizure models (maximal electroshock seizure, pentylenetetrazole-induced and kindling-in...

Influence of Sexuality in Functional Recovery after Spinal Cord Injury in rats

Background: Spinal cord injury (SCI) is a major clinical condition and research is commonly done to find suitable treatment options. However, there are some degrees of spontaneous recovery after SCI and gender is said to be a contributing factor in recovery, but this is controversial. This study was done to compare the effects of sexual dimorphism on spontaneous recovery after spinal cord inj...

Acute Leptin Treatment Enhances Functional Recovery after Spinal Cord Injury

BACKGROUND Spinal cord injury is a major cause of long-term disability and has no current clinically accepted treatment. Leptin, an adipocyte-derived hormone, is best known as a regulator of food intake and energy expenditure. Interestingly, several studies have demonstrated that leptin has significant effects on proliferation and cell survival in different neuropathologies. Here, we sought to ...
